이화여자대학교 질병생리학 연구실(Ewha Nutritional Pathophysiology Laboratory, Ewha NPPL)은 분자생물학적인 방법과 빅데이터를 기반으로 질병의 발생과 예방, 건강기능식품의 안전성 평가 등의 연구를 진행하고 있습니다.


1. Molecular nutrition

- 간질환 예방 및 조절 기전에 관한 연구

간은 인체의 영양소 대사와 독성 물질 제거 등 매우 중요한 기능을 담당하고 있습니다. 간암은 ‘침묵의 암’이라고 불릴 정도로 손상 정도에 비해 증상이 없는 질병 중 하나입니다. 자각증세가 심하지 않아 조기진단에 어려움이 있고 치료 시기를 놓질 경우 좋지 않은 예후를 보이게 됩니다.

본 연구실에서는 간 건강과 밀접한 관련이 있는 것으로 알려진 Prohibitin1 (PHB1)의 발현과 간 질환의 발생 기전을 밝히는데 주력하고 있습니다.  

- 간 오가노이드 개발 및 활용 연구

오가노이드란 인체 장기의 구조적 및 기능적 특성을 모방할 수 있는 3차원적 세포 배양 시스템을 말합니다. 전통적인 세포 배양 시스템은 in vivo 환경을 완벽하게 모방할 수 없으며, 동물실험은 경제적, 윤리적인 한계점을 가지고 있습니다. 따라서 간 오가노이드 구축은 2D culture의 제한점을 극복하고, 동물실험을 최소화하여 in vivo와 유사한 환경을 조성할 수 있다는 점에서 큰 이점이 있습니다.

본 연구실에서는 간 오가노이드를 이용하여 간 질환의 발생 기전 및 예측 바이오마커 개발 연구를 진행하고 있습니다. 또한 간 오가노이드를 이용한 건강기능식품의 안전성 및 독성 예측∙평가 시스템 개발하기 위한 연구를 진행하고 있습니다. 이는 기존에 안전성 및 독성 평가에 이용되고 있던 세포 주 실험과 동물실험을 대체하여 안전하게 인체에 적용할 수 있도록 하는 시스템 개발이 될 것이라 기대하고 있습니다.

- 황 함유 아미노산의 간 보호 효과 구명

황 함유 아미노산 및 그 대사체는 methionine, cysteine과 함께 그 대사과정에서 생성되는 대사물로 S-adenosyl methionine (SAMe), taurine, glutathione과 같은 물질을 지칭합니다. 이 물질들은 체내에서 methyl group 전달 및 항산화 기능을 하는 것으로 알려져 있습니다. 항산화 기전은 간이 손상되었을 때 간을 보호하는 중요한 역할을 담당합니다. 황 함유 아미노산의 대사과정 중 생성되는 glutathione은 강력한 항산화 작용을 통해 간기능 회복 및 간 손상 예방에 효과가 있는 것으로 알려져 있습니다.

본 연구실에서는 glutathione의 전구체인 SAMe, cysteine, taurine과 같은 황 함유 아미노산 및 그 대사체와 대사과정에 도움을 주는 betaine 같은 물질의 섭취가 간 손상 및 간암에서 간 보호 효과를 낼 것으로 예상하고 관련 메커니즘에 대한 연구를 진행하고 있습니다.



2. Data science base nutrition

- 다중 오믹스 분석(Multi-Omics Analysis) 기반 간질환 예측 바이오마커 개발

다중 오믹스 분석은 유전체(genome), 전사체(Transcriptome), 단백체(Proteome), 대사체(Metabolome) 등 다양한 분자 수준에서 생성된 여러 데이터들의 종합적인 분석을 의미합니다. 여러 종류의 오믹스를 동시 분석하고 종합하면 질병의 발생을 예측하거나 진단할 수 있습니다.

본 연구실에서는 Prohibitin1 발현 수준에 따라 변화하는 물질들의 변화를 확인하고 이를 통한 간질환의 발생 기전에 대한 연구를 진행하고 있습니다. 나아가 이러한 물질들을 통해 간 질환 민감성 (liver disease susceptibility)을 예측할 수 있는 바이오마커 개발 연구를 진행하고 있습니다.

- 빅데이터 기반 개인 건강기능식품 안전성 평가

국민의 건강기능식품에 대한 수요가 증가함에 따라 건강기능식품의 중복 및 병용섭취가 이어지고 있습니다. 현재 건강기능식품의 중복/병용 섭취로 인한 안전성 평가방법이 부재한 상황이며, 다양한 식품 및 독성 등 데이터베이스들이 통합되어 있지 않습니다.

본 연구실에서는 대국민에게 균형 있는 건강기능식품 섭취의 방향성을 제공할 수 있도록 건강기능식품 빅데이터를 구축하고, 이를 기반으로 건강기능식품 중복/병용 섭취에 따른 부작용 예측 알고리즘을 개발하여 관련 시스템을 구축 및 보급하는 연구를 계획하고 있습니다.

 

3. 건강기능식품 규제정책

융복합 건강기능식품이란 건강기능식품과 일반식품을 일체형으로 만들어 동시에 섭취가 가능하도록 편의성을 높인 제품을 뜻합니다. 융복합 건강기능식품의 관심이 증가함에 따라 많은 업체에서 융복합 건강기능식품을 만들기 위해 노력하고 있습니다. 따라서 안전하게 운영될 수 있도록 위생교육, 안전점검, 컨설팅 등 지원이 필요합니다.

본 연구실에서는 융복합 건강기능식품 제조업소를 대상으로 점검 및 컨설팅을 진행하여 기준 규격 및 개정안 마련 연구를 진행하고 있습니다.
